The American speedster recovered from her poor start to glide past the rest of the field, but fell short to Jackson's supremacy, crossing the finish line first.
Gabrielle Thomas ran the fastest overall time in semifinal 1, clocking 21.97s ahead of Great Britain's Dina Asher-Smith in 22.28s.
Meanwhile, Julien Alfred continued with her impressive form from the NCAA season, winning the second heat of the semifinal in 22.17s, with Daryll Neita clocking a Personal Best of 22.21s for the second automatic qualification.
Jackson and Thomas are favourites for the title, but Richardson may spring another surprise just like she did in the 100m.
